Nicotinamide riboside (NR) and its mononucleotide form (therefore without ribose and with a single nucleotide) are two molecules that have been part of my interests for several years. For all practical purposes, remember that these are derivatives of vitamin B3.

Nicotinamide Riboside increases energy release, even in the most trained athletes

Nicotinamide riboside and the mononucleotide form are two enzymatic cofactors that occur before NAD / H, the enzyme essential for the metabolism of energy from energy substrates. As you know, NAD + is a coenzyme frequently found at the cellular level, especially at the level of the Krebs cycle for example. I wondered if the NR could still have affected my energy level. If nicotinamide riboside improves energy efficiency as one would expect, this molecule could be of interest to athletes. But since I spend more energy than a sedentary one, I really didn’t expect much. The first effects felt by NR mainly concern the availability of energy during the day, a feeling of increased natural energy, with a marked decrease in fatigue. This is already a very nice thing, but it has nothing to do with taking stimulants either. After about ten days, the effect I hadn’t expected at all from NR started to kick in. The feeling of exhaustion of the muscle fibers and the congestion I felt had increased to the point that I felt very strongly that I had returned to my first strength training 20 years ago. So what to expect from nicotinamide riboside and mononucleotide?

First of all, is to tell yourself that if you are over 40, it is certainly the most useful dietary supplement you could give to your body, whether you are sedentary or athletic. And if even at this advanced age you still have some savvy, you will also think that these two molecules should get along well with Coenzyme Q10, PQQ and resveratrol (obviously). Regarding the nicotinamide mononucleotide, I have also tested it but in my experience, it does not offer the same efficiency at the anaerobic alactic level as the riboside form whereas it is closer to NAD + than the form with the ribose. So if you do strength sport, try NR rather than NMN. If this is the energy you are targeting, Nicotinamide Mononucleotide may be more suitable. At least that’s what comes from my personal experience.

However, the latter is more difficult to access than the riboside form and is currently not patented. This means that you cannot be sure of the quality of the product if you take nicotinamide mononucleotide, unless you can be shown a test from an independent laboratory. But even so, you might still have doubts about the quality of the supplement. According to figures and studies, nicotinamide mononucleotide is four times more effective at restoring NAD + levels than riboside. However, it remains horribly expensive (around $ 1000 / gram) because it is difficult to produce in quantity. Some laboratories already offer it; that is, they must have benefited from a favorable rate on the quantity purchased. This is particularly the case with nicotinamide mononucleotide from Rev Genetics, which I tested for 9 months but the dosage remains modest (50 mg / day and 75 mg for the last 3 months). The feeling of energy is there but oddly enough, it is more felt on the endurance.

On the other hand, the MNM Pure from Alive by Nature seems to me to be reliable. The daily dosage of 125 mg / day is significantly more significant. Again, endurance increases with an ability to run longer, much like regaining your running skills that you had when you were a little younger. The effects were however very slow to be felt. But other than this significant increase in daily stamina and energy, there was nothing more to report. Ultimately, compared to my experience, I would tend to believe that NR would be more effective on the release of energy in anaerobic alactic (ATP / phosphocreatine then glycolysis) while the nicotinamide mononucleotide would somehow promote oxidative phosphorylation. However, there are still no scientific studies specific enough to determine this.

The issue of cellular energy isn’t the only factor to consider when it comes to longevity

Then, we must also say to ourselves that although NR and nicotinamide mononucleotide can positively influence longevity by stimulating the release of cellular energy, it is not the only determining factor of organic longevity – far from it! – to be taken into account when it comes to reducing aging. To this end, I would invite you instead to read Aubrey de Gray’s main popularization book “Ending Aging”. This isolates seven main causes of aging among which he cites nuclear and epigenetic mutations, mitochondrial mutations, intracellular waste, extracellular waste, cell loss, cell senescence and extracellular connectors (binding proteins). Still, the main factor determining whether a cell is alive is that it produces energy. Let’s say that the complexity of the metabolism does not allow us to reduce the question of aging to a few molecules, unfortunately… The reality is much, but really much more complex.

Clinical information regarding nicotinamide mononucleotide and riboside begins to accumulate on the issue of human longevity

From there you understand the paradox but also that nicotinamide, riboside or mononucleotide, if it helps you generate a little more cellular energy, with a better yield, is not a sufficient guarantee for a very greatly increased longevity, at least like the one targeted by researchers. Remarkably, NMN effectively attenuated the physiological decline associated with age in mice without obvious toxicity or adverse effects.

Nicotinamide mononucleotide suppressed age-associated weight gain, energy metabolism was improved, frequency of physical activity increased, with improved insulin sensitivity and plasma lipid profile, improved eyesight and other pathophysiologies. Consistent with these phenotypes, NMN prevented gene expression changes in age-associated diseases in the key metabolism of these organs. It also improved mitochondrial oxidative metabolism as well as mitonuclear protein imbalance in skeletal muscle. These effects of NMN highlight the preventive and therapeutic potential of NAD + intermediates as effective anti-aging interventions in humans.

Nicotinamide mononucleotide and riboside seem to inaugurate a revolution in micronutrition towards increased longevity
